Procedure
Slide Test
1. Place one drop each of Anti-A, Anti-B, Anti-D, and Anti-AB reagents on a clean, dry slide.
2. Add one drop of whole blood to each reagent and mix thoroughly with an applicator stick.
3. Gently rock the slide back and forth.
4. Observe for agglutination within 2 minutes.
Tube Test
1. Prepare a 5% suspension of red blood cells in isotonic saline.
2. Add one drop of each reagent (Anti-A, Anti-B, Anti-D, Anti-AB) into labeled tubes.
3. Add one drop of the cell suspension to each tube and mix well.
4. Centrifuge for 1-2 minutes at 1000 RPM or incubate at room temperature for 20-30 minutes.
5. Gently dislodge the cell button and observe for agglutination.
Interpretation of Results
- Agglutination: Indicates the presence of corresponding antigens (A, B, AB, or Rho).
- No Agglutination: Indicates the absence of corresponding antigens.
Sample Collection and Storage
- Collect whole blood with anticoagulant.
- If testing is delayed, store the sample at 2-8°C.
